A loxP site was inserted upstream of the gene and an FRT site flanked neomycin resistance gene cassette and a second loxP site were inserted downstream. The gene and selection cassette were deleted in the germline through cre-mediated recombination by crossing mice carying the floxed allele with Zp3-cre allele carying mice. This creates a knockout allele. (J:271669)
