Exon 2 was flanked by loxP sites, with one loxP site positioned inside the 3' UTR of exon 2. A FRT-flanked neomycin cassette that was inserted between the first loxP site and exon 2 was removed via flp-mediated recombination. Cre-mediated recombination deleted the exon 2 open reading frame. Immunohistochemistry confirmed the absence of protein in hemizygous male embryos at E10.5. (J:271028)
