A targeting vector replacing exons 2-5 with a lacZ cassette and a loxP-flanked neomycin resistance cassette was obtained from Velocigene. Cre-mediated recombination removed the neomycin cassette leaving behind the inserted lacZ reporter sequence. Successful deletion was confirmed by PCR, immunoblotting of gluteus lysates, and mass spectrometry using an in vivo stable isotope labeling by amino acids in cell culture (SILAC) approach. (J:270838)
