A loxP-flanked neomycin cassette replaced exon 2 of the gene, which encodes the N-terminal part of the epidermal growth factor-like domain in the encoded protein. Deletion of exon 2 introduces a frameshift mutation in the truncated transcript. Western blot analysis of brain lysates showed that no detectable encoded protein was present in homozygous mice.
