The transgene contains the cDNA for a mutated form of the rat cAMP responsive element binding protein 1 downstream of the tetracycline-responsive element. The cDNA has A>G and A>C point mutations changing codon 304 (p.K304A) from lysine to alanine, thereby inactivating or severely reducing the peptide's capacity to bind to the cAMP response element (CRE) on the DNA, without affecting dimerization with wild-type peptide. The cDNA sequence had been previously modified to change codons Cys300, Cys310, and Cys337 to serine to improve solubility. (J:76362)
