A construct, containing an FRT flanked cassette, encoding a splice-acceptor site, lacZ gene and neo resistance gene, and a loxP site, was inserted in intron 3 and a second loxP site was inserted in intron 4 via homologous recombination. Western blot analysis of lysates of MEFs from homozygous mice indicated the absence of protein expression. (J:268910)
