A floxed puromycin with a splice acceptor, four SV40 polyadenylation sequences and splice donor was inserted upstream of exon 8. Exon 9 was modified with a C to G point mutation to induce the amino acid substitution of arginine for proline at position 286 (P286R). This mutation is present in human cancers and leads to error-prone DNA replication. (J:266324)
