The critical C and G in the two Trp53 response elements in intron 1 were mutated. These mutations abolish the activity of the P2 promoter. An inverted floxed neomycin resistance cassette was inserted downstream of exon 3. Cre-mediated recombination removed the selection cassette. (J:201057)
