A bidirectional tetracycline/doxycycline- (tet/dox-) responsive promoter drives expression in one direction of the E. coli lacZ (beta-galactosidase) gene, and in the other direction of a LINE1 retrotransposition reporter cassette ("ORFeus"). The ORFeus cassette includes 1) the codon-optimized open reading frames (ORF1 and ORF2) of a human LINE1 (long interspersed nuclear element 1), encoding the proteins required for its retrotransposition; and 2) a green fluorescent protein (GFP) reporter gene, interrupted by an intron, inserted in reverse orientation into the LINE1 3'-untranslated region (3'-UTR). lacZ expression serves as a reporter of tet/dox-induced transcription, and excision of the intron from the ORFeus transcript permits expression of GFP from a retrotransposed copy.
