The targeting vector was designed to insert a loxP site upstream of exon 2, and a frt-flanked and loxP-flanked neomycin selection cassette downstream of exon 5. Flp-mediated recombination removed the neo cassette and left exons 2 through 5 floxed. (J:269891)
