A PAC containing both variants (p16-Ink4a and p19-Arf) of the mouse Cdkn2a gene, plus the Cdkn2b gene, was engineered to replace the upstream Arf variant exon 1-beta with a chimeric gene. The chimeric gene was inserted at the translation start codon of the host gene and consists of the human heparin-binding EGF-like growth factor gene, with p.I117V and p.L148V mutations, linked to the firelfly luciferace gene via sequence for the self-cleaving picornavirus-derived 2A peptide. The mutations in the human gene disable the growth factor function without affecting the diphtheria toxin receptor function. Thus, cells that express this allele will go into apoptosis when diphtheria toxin (DT) is administerd to the mouse. The PAC was microinjected into pronuclei of C57BL/6J embryos. (J:265488)
