The transgene is designed with a bidirectional tetracycline operator (tetO; also called tetracycline-responsive element (TRE, TetRE) or tet-operator), Bi-L-Tet, located between two cytomegalovirus (CMV) minimal promoters in the opposite orientation. One CMV promoter drives expression of the murine DEK oncogene (DNA binding) gene (Dek) followed by a polyA signal, while the other CMV reporter drives expression of a luciferase (luc) reporter followed by a simian virus 40 (SV40) polyadenylation (polyA) signal.
