A cDNA encoding cre-IRES-EGFPnls (nuclear localized EGFP) as well as an FRT-flanked neomycin cassette was recombined in-frame with the Ly6a ATG start codon (exon 2) via homologous recombination. Resultant mice were crossed with Flpe mice to delete the neomycin cassette. Though present in the targeting construct, IRES-driven expression of EGFPnls is not observed. Expression of Ly6a is eliminated by the targeted allele. (J:264764)
