A targeting construct was designed to replace the stop codon of the growth hormone releasing hormone (Ghrh) gene with a viral P2A oligopeptide, from porcine teschovirus-1, fused to a Cre recombinase gene. A frt-flanked neomycin resistance (neo) cassette inserted downstream. The construct was electroporated into R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred with B6.129S4-Gt(ROSA)26Sor/J mice to remove the neo cassette. (J:264547)
