A mature B cell, where the the immunoglobulin heavy chain complex had undergone Igh-D to Ighj1 recombination and subsequently an IgE class-switch, was isolated and reprogrammed to an iPSC (induced pluripotent stem cell). This iPSC was then used to create a homozygous mouse line where all cells contain this non-productive allele. The mouse from which the B cells were isolated is a reprogrammable mouse containing the Col1a1tm1(tetO-Pou5f1,-Klf4,-Sox2,-Myc)Hoch and Gt(ROSA)26Sortm1(rtTA*M2)Jae alleles which allow for the reprogramming to be induced by doxycycline. Sequencing of the DJ junction could not distinguish between the recombination involving DSP2.7 (Ighd2-11*02) or DSP2.8 (Ighd2-10*02). (J:252910)
