The human bacterial artificial chromosome (BAC) RP11-237K15 containing the entire TREM2 gene with adjacent 50 kb + flanking regions, and the TREML1, TREML2, and TREML4 genes, was modified by deleting the TREML1, TREML2, and TREML4 genes. The modifications to the BAC were as follows: exons 1 through 3 and the proximal promoter region (most of the protein coding sequence) were deleted in TREML4; the proximal promoter region as well as exons 1, 2, 5 and 6 (80% of the protein coding sequence) were deleted in TREML1; and exons 2 and 3 of the TREML2 gene were deleted (resulting in a frameshift with early stop codon in exon 4). Founder line A, which carries 1 or 2 copies of the transgene (transgene copy number estimated by genomic qPCR) was subsequently established. (J:262250)
