The transgene comprises the following elements: the mouse nestin promoter, a modified reverse tetracycline-regulated transactivator gene, an IRES, the EGFP fluorescent marker gene, a poly(A) signal and nestin gene intron 2. The intron directs the expression of the two inserted genes to neuronal precursor cells only, preventing the expression in somites that would have occurred if under control of the promoter only. When mice carying this transgene were crossed with mice carying a tetO controlled transgene (Tg(tetO-cre)1Jaw), the resulting double-transgenic offspring showed doxycycline-controlled expression of the second transgene. When further crossed with mice carying a conditional reporter allele (Gt(ROSA)26Sortm1Sor), triple-transgenic mice showed doxycycline-inducible expression of the conditional transgene. (J:97312)
