The human U6 small nuclear RNA gene promoter was used to drive expression of a mouse anti-Cdk5 shRNA. The construct was microinjected into the male pronuclei of C57BL/6J zygotes. Three founders were generated. Immunohistochemical and Western blot analyses confirmed that protein expression was decreased in the skin of knockdown mice relative to wild-type controls.
