The transgene comprises the following elements: the mouse cardiac myosin heavy chain promoter, DNA sequence coding for amino-acids 2-118 of the human transferrin receptor (includes transmembrane sequence), DNA sequence coding for amino-acids 139-387 of the chicken ovalbumin gene, and, from the pcDNA3.1V5-His6-TOPO plasmid, V5 epitope sequence, His6 tag sequence, stop codon, and bovine growth hormone polyA signal. RT-PCR, ELISA and immunofluorescence experiments confirmed exclusive expression in myocardium. The construct was microinjected into embryonic stem cells. Mice carying this transgene will express a membrane-bound chimeric protein with the ovalbumin peptide exposed extracellularly. The ovalbumin peptide includes the two sequences that are recognised by the T cell receptors expressed by transgenes Tg(TcraTcrb)1100Mjb (OT-I) and Tg(TcraTcrb)425Cbn (OT-II) respectively.
