Tg(KRT14)1Efu

The transgene comprises the following elements, all from human keratin 14: upstream sequence (from the HindIII site 2.5 kb upstream of the transcription start site) including the promoter, the cDNA with the antigenic portion of neuropeptide tachykinin 1 (substance P) inserted in-frame 18 bp upstream of the stop codon, and 3' UTR sequence including poly(A) signal (up to the EcoRI site 700 bp downstream of poly(A) signal). The construct was micro-injected into the male pronuclei of fertilized oocytes. Immunohistological analysis using substance P antibody shows expression in the basal layer of the epidermis and in the outer root sheath cells of the hair follicles. Line 1 showed consistent (non-chimeric) expression of the transgene. (J:113642)
