A loxP site was inserted 40 bp upstream of the coding region of the target gene, and a second loxP site downstream. An FRT site flanked neomycin resistance gene cassette was inserted between the target gene and the downstream loxP site. The neo cassette was subsequently removed through flp-mediated recombination, leaving a conditional-ready allele with the gene floxed. (J:262024)
