The transgene comprises a 15.9-kb human genomic DNA fragment containing the wild-type allele of the rhodopsin gene with 4.8 kb of DNA upstream of the start codon and 6.2 kb of DNA downstream of the termination codon. Densitometric analysis of transgene-derived human and endogenous mouse rhodopsin mRNA-derived bands on autoradiographs of polyacrylamide gel-separated [alpha-32P]-dCTP-labeled RT-PCR products of retinal mRNA from heterozygous transgenic mice revealed a human:mouse transcript ratio of 1:1.
