A fragment of mouse genomic sequence from 453 bp upstream of the transcription start site of the Col11a2 gene to 380 bp downstream (the former being promoter and the latter being 5' UTR sequence), was cloned upstream of a construct containing SV40 intronic sequence and the mouse Cilp gene with influenza virus hemagglutinin (HA) epitope sequence in-frame with the 3' end. A 2.3 kb DNA fragment representing most of the first intron of Col11a2 was inserted downstream of this. This construct was microinjected into the pronuclei of C57BL/6 fertilized eggs. (J:219126)
