This knockout-first reporter allele with conditional potential was generated by electroporation of embryonic stem cells with the AsiSI-linearized targeting vector HTGRS6004_A_H09, obtained from KOMP. The vector introduced a FRT site; the mouse engrailed 2 (En2) intron, splice acceptor and exon; an internal ribosomal entry site (IRES); the lacZ coding sequence with a polyadenylation signal (p[A]); a loxP site; a neomycin resistance cassette with the human beta-actin promoter and a p[a]; a second FRT site; and a second loxP site into intron 3 and a third loxP site into exon 5.
