The targeting vector contains (5' to 3'): CAG (cytomegalovirus early enhancer/chicken beta-actin) promoter, an FRT site, loxP-STOP-loxP cassette, Salsa6f (see below), WPRE, bGHpA, AttB site, an FRT site, neomycin selection cassette, AttP site (using the Addgene Plasmid #34883); inserted into the Gt(ROSA)26Sor locus. PhiC31-mediated recombination removed the AttB/AttP-flanked sequence. Salsa6f contains tdTomato linked to GCaMP6f by a V5 epitope tag. (J:254963)
