The transgenic construct consists of the tetO, tetracycline-responsive regulatory element (from pTRE-tight), cDNA encoding the Drosophila AstA-R1, or AlstR, allatostatin A receptor 1, gene and a SV40 polyadenylation sequence. The TRE promoter used here is Tet-responsive Ptight; containing a modified Tet response element composed of seven direct 36 bp repeats (each containing the 19 bp tet operator sequence) and a minimal human cytomegalovirus promoter, which lacks the enhancer that is part of the complete CMV promoter. Consequently, Ptight is silent in the absence of TetR or rTetR binding to tetO. (J:261666)
