A floxed mini-gene containing exons 4-7 and a BGH poly-A signal were inserted upstream of exon 4, M396A and F398A mutations were inserted in exon 4, and an F3 site flanked pgk-hygro cassette was inserted in upstream of the mutated exon 4 via homologous recombination. Targeting was done in ES cells carrying Birc3tm1Rbr . Flp and cre mediated recombination removed the pgk-hygro cassette and floxed mini-gene. (J:260804)
