A human BAC (BAC H0193H09 from the CEPH) carrying RHD gene was used to make transgenic mice. Transcripts from the human gene are detected, but no protein expression was detected with a panel of anti-RhD monoclonal antibodies recognizing different epitopes or a polyclonal anti-RhD. Human RhD protein is not expressed in mouse erythroid cells in the absence of human RHAG despite the presence of murine Rhag. In the presence of human RHAG, a low level of human RHD is detectable.
