A TALEN-mediated mutation was produced by targeting exon 4, which contains the initiation codon ATG. A 43-bp deletion from this genomic region was confirmed by sequencing, resulting in a frameshift mutation with a premature stop codon. RT-PCR analysis confirmed the absence of mRNA expression in whole brain lysates without the hippocampus as well as in hippocampus only lysates from homozygous mutant mice. (J:257371)
