A loxP site was inserted into intron 3 and an FRT site flanked neomycin resistance gene cassette and a second loxP site into intron 13. The neo cassette was removed through subsequent flp-mediated recombination. Exons 4-13 were deleted through subsequent cre-mediated recombination. Exons 8-13 code for the kinase domain. qRT-PCR and western blots confirmed the absence of expression from this allele. (J:226871)
