The transgene was constructed using BAC recombineering with galK (galactose kinase) selection. BAC clone RP97.23H11 carries the TH2 locus-control region, Il4, Il13 and Kif3a. The ATG start codon of the Il4 gene is targeted with galK, galK is then targeted with AmCyan (Anemonia majano cyan fluorescent protein) sequence. Similarly, the ATG start codon of the Il13 gene is targeted with de-stabilized DsRed (DsRed-DR) via galK selection. AmCyan and DsRed expression is correlated with Il4 and Il3 expression, respectively. (J:197353)
