A cassette containing a Gateway sequence, FRT site, splice acceptor, IRES, beta-gal, LoxP site, neo cassette, FRT site, loxP site and a final Gateway sequence was inserted upstream of exon 5 and a third loxP site was inserted downstream of exon 5 via homologous recombination. Flp-mediated recombination removed most of the cassette, leaving exon 5 floxed. (J:256762)
