The initial targeting event inserted loxP site 3 to the endogenous exons 14-15, a synthetic poly(A) site preceding a FRT-Neo-FRT cassette, a second loxP site, and a second copy of exon 14 (the last coding exon of the gene) fused to AcGFP sequence. FLPmediated recombination removed the neo selection cassette, and Cre-mediated recombination removed the endogenous exons 14 and 15, leaving the second copy of exon 14 (the last coding exon of the gene) fused to AcGFP sequence.
