An FRT-flanked neomycin resistance cassette with a 5' loxP site was inserted upstream of exon 21. An F3-flanked puromycin resistance cassette with a 3' loxP site was inserted downstream of exon 21. Exon 22 was replaced with a modified exon in which a nucleotide substitution results in the amino acid substitution of alanine for aspartic acid at position 761 (D761A). Flp-mediated recombination removed both selection cassette. (J:255862)
