The nuclear localization sequence (NLS)-cre-poly A-FRT-amp-FRT cassette was inserted using Red/ET recombination techniques into the translation initiation site located in the 2nd exon of the mouse Trpc4 gene in a BAC clone RP24-67D9. Resistant clones were selected and confirmed for appropriate homologous recombination, after which amp was removed prior to the microinjection. The resulting vector was purified, linearized and microinjected into pronuclei of fertilized C57BL/6J eggs. The founder G was crossed with C57BL6J mice and the subline 383 was crossed with C57BL6J mice. (J:255267)
