This allele is generated using somatic cell nuclear transfer (SCNT). Individual (C57BL/6 x 129)F1 donor nuclei from iNKT (invariant natural killer T) cells were isolated and used to replace the mitotic spindle from mouse oocytes. Cells are developed to the blastocyst stage and used to generate embryonic stem (ES) cells. ES cells were injected into recipient blastocysts, and blastocysts implanted into pseudopregnant females. The iNKT cells were selected to carry a rearranged invariant Tcra V14-J18 segment. (J:254145)
