Codon 455 GAA encoding glutamic acid in exon 4 was changed to AAA encoding lysine (p.E460K) and an FRT site flanked neomycin selection cassette was inserted into intron 3. This mutation is equivalent to the human p.E455K mutation associated with dilated cardiomyopathy (DCM). Western blot analysis confirmed expression of the protein in the hearts comparable to BAG3 protein levels in wild-type controls. (J:246352)
