A construct comprising the following elements was inserted into the host locus: CAG promoter, a loxP site, a neomycin resistance gene cassette, a STOP cassette, a loxP site, the mouse gene with a 3' fusion of sequence coding for a hemagglutinin (HA) tag, an FRT site, IRES sequence, the eGFP fluorescent reporter gene, an FRT site, and poly(A) features. The inserted mouse gene and the reporter gene will only be expressed after removal of the neo-STOP cassette through cre-mediated recombination. The reporter gene can be removed through flp-mediated recombination. (J:243119)
