The targeting vector is designed to insert, by site-directed mutagenesis, a point mutation (GAC to GCC) at nucleotide 419 followed by an FRT-flanked neomycin cassette downstream of exon 4. The mutation results in an alteration of the corresponding amino acid from aspartic acid to alanine (D140A) at codon 140. Flp-mediated recombination removed the FRT-flanked neo cassette. The mutation does not affect peroxidase activity, but does eliminate phospholipase A2 activity. (J:239138)
