Sequences encoding for the simian diphtheria toxin receptor (DTR) fused to EGFP and bGH polyA sequences downstream of a double-stop (ds) cassette consisting of a floxed STOP signal and an frt flanked nuclear bata-galactosidase (nlsLacZ) and STOP signal was inserted into Mapt locus. Endogenous Mapt promoter drives expression of nlsLacZ or DTR/EGFP fusion protein. Cre mediated removal of the first STOP signal is required for expression of nlsLacZ. Cre and flp mediated recombination are required to remove both STOP signals to allow for expression of the DTR/EGFP fusion protein. (J:227180)
