An FRT-lacZ-loxP-Neomycin/pA-FRT-loxP cassette was placed in intron 3 and a loxP site was introduced to intron 4 via homologous recombination. Transient infection of the ES cells with a plasmid expressing FlpE (pCAGGS-FlpE-PURO) excised the lacZ and neomycin components, leaving exon 4 flanked by loxP sites. (J:235710)
