A loxP site flanked neomycin resistance gene cassette was inserted. The part of the gene coding for the compaction region, one of the active domains of the protein, was replaced with synthetic sequence where 23 selected lysine and arginine codons had been changed to alanine codons. The neo cassette was removed through subsequent cre-mediated recombination. The result is an inactive protein. (J:242063)
