The coding sequence of Sox10 in a phage artificial chromosome (PAC; RP21-529-16) was replaced with an icre/ERT2 cassette by homologous recombination in bacteria. The targeting vector also contained an Frt-flanked chloramphenicol resistance gene, which was removed by transient activation of Flp in bacteria. A construct was generated that encodes tamoxifen-inducible cre under control of the Sox10 regulatory sequences. The purified targeting construct was used to generate transgenic mice. These animals were crossed to ROSA26-EYFP reporter animals for analysis. Data for line 26 (SER26, [S]ox10::icre[ER]T2) was presented and was also confirmed by analysis of lines 68, 87, and 93.
