A construct consisting of an FRT site, neo cassette and an FRT, lox2272, lox5171 flanked inverted splice acceptor site-GFP-polyA cassette was inserted in intron 1. Flp mediated recombination removed the neo cassette leaving the GFP cassette unchanged. The lox sites are arranged to allow for cre mediated inversion of the GFP cassette that is fixed after 2 successive cre mediated recombination events. Inversion of the GFP cassette traps the gene. The FRT sites are arranged to allow for Flp mediated deletion of the GFP cassette restoring gene function. (J:228038)
