A loxP site was inserted in intron 13 and IRES-EGFP-Luciferase cassette, FRT flanked neo cassette and loxP site were inserted after the stop codon in exon 17. Flp mediated recombination removed the neo cassette. In heterozygous and homozygous MEFs, mRNA expression is reduced to about 70% and 30% of wild-type levels. (J:227638)
