A targeting construct was designed to insert a Cre Recombinase gene and a zsGreen (enhanced green fluorescence protein) just upstream of the 3'UTR of the angiotensin II receptor, type 1a (Agtr1a) gene. Viral P2A oligopeptides, from porcine teschovirus-1, were inserted upstream and downstream of the cre gene to enable ribosomal skipping. A frt-flanked neomycin resistance (neo) cassette was placed downstream of the 3'UTR. This construct was electroporated into C57BL/6J embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and the resulting chimeric mice were bred to FLP (FLPo) recombinase expressing mice to delete the neo cassette. (J:240544)
