The targeting vector is designed to insert a single loxP site upstream of exon 3 and a single loxP site downstream of exon 5 of the gene. An FRT-flanked neomycin resistance (neo) cassette is inserted between exons 2 and 3. The neo cassette was deleted by transiently transfection of an expression vector of flp recombinase expression plasmid in the ES cells. (J:101977)
