A bacterial artificial chromosome (BAC) library was used to obtain a 110 kb BAC containing the entire human glyceraldehyde-3-phosphate dehydrogenase (GAPDH) gene. A loxP-flanked STOP signal, followed by a human catalase (CAT) gene, was inserted into this BAC to replace exons 3-7 of GAPDH. The initiating methionine of CAT was deleted, and the first 25 amino acids of the ornithine transcarbamylase (Otc) leader sequence were added to the amino terminus to target this protein product to mitochondria.
