The CAG promoter drives the expression of floxed lacZ followed by three polyadenylation (pA) signals. Following this is the Nos1ap open reading frame (ORF, NM_001109985) including start and stop codons; this is then followed by an IRES-puromycin-pA cassette. In the presence of cre recombinase, the floxed lacZ-polyA cassette is deleted, leaving Nos1ap under the control of the strong constitutive enhancer-promoter element. This leads to Nos1ap overexpression in the tissues in which cre recombinase is expressed. Lines 112, 314 and 333 were generated. (J:232078)
