The targeting vector is designed to replace the stop codon in exon 18 of the Fgfr1 gene with a cassette containing a human HIST1H2BB gene [histone 1 H2bb] sequence, a viral 2A oligopeptide (T2A) that mediates ribosomal skipping, a cerulean variant of green fluorescent protein (GFP) sequence, and a new stop codon. An FRT-flanked neomycin resistance (neo) cassette was inserted downstream of the new stop codon. Flp-mediated recombination removed the FRT-flanked neo cassette. (J:241560)
